First is the spores are similar to the corticeps that inspired The Last of Us. Then there is a parasite that infects some animals and thats what you thought of that makes birds eat them and the eggs of said parasite are then put into the fecal matter of the bird to begin the life cycle anew. But the comparison is there. Corticeps actually make ants climb up a high area to disperse spores across the other ants, and that cycle continues. That is what inspired the last of us and it seems to be the basic inspiration for the smile spores we see here. Though thats just based on the video of the first test subjects.
Edit*it actually takes 2 spores that are compatible to start the growth of fungus once more, 1 is not enough to grow the fungus
Fungal spores are flammable which suggest why they are weak to fire, but they must of built of some resistance, so the only way to ignite them is if a source of direct contact from fire for a period of a couple or even half a second.
So, what I think is that, initially, this was meant to represent heavier topics like depression or other mental illnesses, where people just hide behind their smile, acting like they don't have anything wrong with them, like they are doing fine, with nothing wrong with them, laughing and smiling just like everyone else. Eventually, it seems to go away from this kind of topic, and towards a more usual horror situation.

It's not analog horror, but you should definitely watch Marble Hornets! If you like Slenderman, and Creepypastas, you'll like Marble Hornets. It's known to feel real to the viewer due to the filter on information we, the viewer receive.

Fun fact about Acetylcholine 6:21 :
There is an amoeba which loves acetylcholine it is called naegleria fowleri should any water get up high inside your nose there is a risk of the amoeba finding your olfactory bulb, it then follows your olfactory nerves up to your brain, it sees your nerves which have a lot of acetylcholine and immedietly begins attacking and eating your brain, most patients died.
If you're looking for other analog horror-style stuff to react to, I'd like to recommend a couple people to check out: VibingLeaf, who has their own Sonic.exe series, as well as retakes of various creepypastas, including things like Red Mist and Mouse.avi, and anormalshark, who also has a series revolving around a character in the Sonic.exe pantheon, No More Innocence, or NMI, there's not many videos out in that series, but they're quite well made.
