If poison works the same way that it does in Backpack Hero then you're better off stacking the poison as that will end up doing more damage.
In the spirit details you see what seems to be their type in blue but also what types they're weak to in red, which doesn't correlate directly with their type. Although if you're going to knock one out, no point poisoning it first.
